About Scientist, TCR & pHLA Engineering & Production - Waltham, MA

Keyway TM TCR Discovery group is looking for a highly motivated and enthusiastic Scientist to join our team in Waltham, MA. This position will focus on T cell receptors (TCRs) and peptide Human Leukocyte Antigen complex (pHLA) engineering and production to support the discovery of novel TCR biotherapeutics. The candidate must have in-depth knowledge of current state-of-the-art T cell receptors engineering and production and be able to apply these practices at the bench. This role will drive technology developments and optimization of pHLA complex and soluble TCR production in prokaryotic and eukaryotic systems to improve TCR drug developability for development as a drug candidate.

Principal Responsibilities

  • Develop prokaryotic and eukaryotic cell-based expression system for pHLA complex and soluble TCR production
  • Lead pHLA and TCR material generation to support discovery campaigns and downstream functional characterization
  • Run bispecific and multispecific functional TCR engineering and production
  • Perform E. coli protein inclusion body extraction, purification, and refolding
  • Perform protein purifications and troubleshoot any issues with purification using AKTA systems
  • Use automation to support soluble TCR reformatting, production, purification, and screening
  • Express and produce selected TCR leads in micro-scale in a high-throughput manner
  • Characterize TCR binder properties in various affinity and activity assays, like ELISA, iQue, FACS, Octet, epitope binning, reporter gene assays
  • Engineer and improve binders by means of affinity maturation
  • Excellent written and oral communication skills are necessary in order to succeed in a cross-functional scientific environment
  • Multitask and accomplish goals under project and team timelines
  • Be generous when others are needy, greedy, or fearful

Required Qualifications and Skills

  • PhD in molecular biology, biochemistry, cell biology, or related field; MS with 4+ years of industry experience; or BS with 6+ years of industry experience; Title will be aligned with experience level
  • Deep understanding of protein chemistry, including analyses, structure, and function
  • Experience purifying and characterizing proteins, ideally antibodies and antibody fragments
  • Experience with biophysical tools: DSC, DSF, DSC, ce-SDS, cIEF, mass spectrometry
  • Proven track record in protein expression, purification, and engineering
  • Skilled at FACS sorting and flow cytometric analysis
  • Experience with phage display, yeast display, mammalian library, and screening are a plus
  • Structure-guided protein engineering is preferred
  • Ability to conduct data analysis, interpret experimental results, and independently draw conclusions
